The standard everyone cites and almost nobody has read
ASME BPE, the Bioprocessing Equipment standard, occupies an interesting position in pharmaceutical manufacturing. It is widely referenced in design specifications, qualification documents, and supplier conversations. It is cited in audits. It appears in tender requirements.
It is also, in most facilities, only partially understood by the people responsible for implementing it day to day. This is not a failing. The standard is extensive, technical, and written primarily for designers and engineers specifying new equipment. The maintenance technician tightening a tri-clamp connection on a Wednesday afternoon was not the primary audience.
But the standard’s requirements land on that technician’s work nonetheless. And the gap between what BPE requires and what actually happens on the floor is where most connection reliability problems originate.
What BPE actually covers
ASME BPE covers the design, materials, fabrication, inspection, and testing of equipment used in bioprocessing, including the hygienic connections that are the focus of this series. The sections most relevant to daily operations address material requirements for product-contact surfaces including seals and gaskets, surface finish requirements that affect cleanability and biofilm risk, assembly requirements covering gasket selection, clamp type, and torque application, dimensions and tolerances ensuring components from different manufacturers are genuinely compatible, and training requirements for personnel assembling bioprocessing connections.
The four assembly requirements in plain English
1. The right gasket material
- Not just any material that meets USP requirements
- The specific material appropriate for the temperature, chemical exposure, and frequency of sterilisation in that connection
- EPDM and silicone are not interchangeable in a high-frequency SIP environment
2. The right clamp type
- Standard tri-clover clamps are not appropriate for all applications
- High-pressure clamps, heavy-duty clamps, and bespoke configurations exist for a reason
- Using a standard clamp where a high-pressure clamp is required is a maintenance liability, not a cost saving
3. A defined torque standard
- BPE is clear that connections should be assembled to a documented specification, not to feel
- This requires knowing the correct torque value for the specific clamp, gasket, and service condition
- The record of that assembly should be retrievable
4. Trained personnel
- The standard specifically addresses personnel competency
- Someone shown how to assemble a connection once, years ago, has not been trained to BPE
- Training means documented instruction, demonstrated competency, and periodic review
What this looks like in practice
- Defined materials documented in SOPs
- Defined clamp types matched to service conditions
- Calibrated torque wrenches with recorded values
- A training programme with documented sign-off
What it is not
- A checklist ticked once at commissioning
- Whatever the previous technician used
- Components sourced from the shelf without reference to the specification
- Training that happened at induction and hasn’t been revisited since
The standard doesn’t require perfection. It requires specificity. Defined materials, defined clamp types, defined torque, defined training. The opposite of whatever works.
Why this matters beyond compliance
ASME BPE is guidance rather than a hard regulatory requirement in most jurisdictions. But its requirements map directly onto GMP expectations around process control, personnel training, and documentation. An investigator asking about a connection failure will ask the same questions BPE answers: what was the specification, who assembled it, how, and how do you know?
The facility that can answer those questions clearly is in a fundamentally different position, operationally and from an audit perspective, than one whose answer is it’s how we’ve always done it.

- Produce a simple torque specification document covering the clamp types and gasket materials in use

The standard was written for designers
The benefits are for the people maintaining the connections on Monday mornings. A site that implements BPE principles practically does not need a major programme. It needs a review of gasket specifications, a clamp audit, a simple torque standard document, and a training session.
